It is mostly a waste of time and effort as there are several distro's available that can be installed to and run from a usb drive without any modification.
The most notable and popular would be ubuntu, I personally run it on 3 different flash drives with no special config necessary. I also have wine (actually crossover)installed and use a few portableapps in ubuntu on a flash drive.

It runs under quemu, the pendrive has similar versions too, if you wnt boot from the stick, then this has not much to do with portable apps then.
