I am running a copy of TiddlyWiki on my portable hard drive. I would love to have the wiki be one of the start tabs on PF. I have tried many versions of the path using a relative address, including many .. and . addresses. Is there a more straightforward way to do this? Or have I just not hit the proper path yet.

You need to use an INI file. You can find an example in FirefoxPortable\Other\FirefoxPortableSource. Read the readme.txt file in there as well for the full details. The setting you want is called LocalHomepage. You can't use it with tabs (multiple homepages), but it will let you use TiddlyWiki with Firefox Portable as a homepage. In fact, this option was added specifically for TiddlyWiki.
