I am using Thuderbird Portable on a USB drive that I take with me on the road and use from multiple computers. The drive letter changes depending on what is in use already on the machine. Does this present any problems with TBP or should it be the same drive letter each time. If so, what do I need to modify to change the drive letter to something else as my original install was F and I would like to make it a less common one like P or something.

That's kinda the point of the portable version. So it can handle changes to drive letters and paths.

Well yes that makes sense, but what about all the references to F:\..... in the prefs.js and any other files that reference the F drive?

Most of it is handled. The old chrome.rdf (if you use 1.0) is rewritten. The component registry is deleted and allowed to be rewritten by Firefox on each launch.
Rather than questioning it... why not just try it and you'll see that it works.
