what file

What file are you talking about? The KeepassPortable.exe or your database?
Most likely reinstalling will fix the problem. All your data will be preserved on upgrade (unless you have put your database inside the KeepassPortable/App folder).

Open It

You need to open KeePass Portable and then use file open to open your database. You can set it to automatically open your last opened file. The KeePassPortable.exe launcher will adjust this selection as your drive letter changes so it will keep opening.

What? Wait a second, Remote PC? Is that via a UNC Path?

You say "When trying to access it on a remote pc …", you're not trying to use it from a UNC path (e.g. \\Server\path\to\share\KeePassPortable) are you? UNC paths are unsupported, if you need to use it over a network, map the Network Share to a Drive Letter.

To map the network share to a drive letter

  1. Right-Click "My Computer" [XP/2k] or "Computer" [Vista/7]
  2. Click "Map Network Drive…"
  3. Choose a Drive letter
  4. Click "Browse" and go find the network share
or type out where it is if you know it (e.g. \\ServerName\Path\to\share)
  5. Check or uncheck "Reconnect at logon" based on your preferences
