When I use the Password Manager to save the password of my E-mail account, it says that the data is stored in a file on my computer. Therefore I get the advise to use a Master Password.
I assume that the file is stored on the USB stick and not on the PC, or else it would be useless ?!
In either case, where is the file stored ?

The passwords are stored in a file in the Thunderbird "profile", on the USB drive. Nothing is left on the PC. Note that the Master Password only protects your passwords and certificates and such, not email that has been downloaded; but that's pretty standard.
The preview doesn't get a "real" date until you submit it... a strange bug, but that's also pretty standard for the preview.
