Can I use the Auto-Update function from Thunderbrid or must I use the Thunderbird PORTABLE setup to bring my TB installation to the newest version

In general, it is never wise to use the auto update in any of the apps here since they will usually just download and install the full local version onto the current PC.
In specific, the Mozilla apps can almost do an update in place, but, due to a bug introduced a few versions ago, that currently is not the case. Mozilla are aware of the problem, and are working on a fix.
So, at the moment, do not use the inbuilt update function in Thunderbird Portable.
