How do these options work, and how much can they speed up PFF?

Chrome isn't really used anymore as 1.5 changed the file layouts. It's still there to maintain compatibiliy with 1.0. Thus, turning it off won't speed launch at all.
Compreg has to be run when the drive letter changes or FF won't work.

So should I turn off compregfix and only turn it on before I use the computer that uses a different drive letter?

I recommend leaving it on. Sooner or later, you'll forget to reenable it when your drive letter changes. But, if you want to, you can disable it and reenable it before running it on a PC with a different drive letter.

Turning off the compreg fix has sped up Fx's speed a lot. It opens in less than 10 seconds now on a PC with USB 2.0 ports. Before, it took 20-30 seconds.
