Yes

In Windows, you just copy it into your local Thunderbird profile folder:
C:\Documents and Settings\[user]\Application Data\Thunderbird\Profiles\????.default
and delete the compreg.dat folder (if there is one).

In Linux, the local profile folder is ~/.thunderbird// or ~/.mozilla-thunderbird. Anything requiring full paths will fail (signatures, some extensions, etc), but everything else should work... in theory. Give it a shot and let us know!

More specific...

To be more specific, you need to copy all the files within:
\PortableThunderbird\Data\profile\

to your local profile directory:
C:\Documents and Settings\[user]\Application Data\Thunderbird\Profiles\rk2wu308.default

You don't copy the profile folder itself into there. Just all the files. And be sure to delete compreg.dat from the local copy after you copy it in. BTW - If you use certain extensions... this copying to the local PC won't work.

Instead of copying files

Sync does it for you.
And I had to give a plug of Steve's software.

Try using PUSS

Try PUSS, from the Liberta Project (http://liberta-project.org/liberta/core/puss), and use the sync feature to copy your profile across.
