I've succesfully set up the above on a USB stick and am wondering about portability of the three applications.
Will I still have full access to all the options I have on the system on which I set them up, since GnuPG is of course installed on the local drive?

You need to follow the instructions outlined here:

Then you'll have portable everything.

Do you know, if I had Enigmail already installed, then installed the GPG for Portable Apps, do I need to uninstall and reinstall Enigmail or will the original installation be ok?

Think I've just answered my own question as I can see in TBP settings / Enigmail settings that it has automatically found and overridden the default location for gpg.exe
As you've found, our Thunderbird Portable launcher automatically sets it up for you once GPG portable is added on

I am using portableapps for a couple of years now. And until recently I was using TB version 2 and enigmal/GPG/OpenPGP.
Today I updated to TB 3.0.4. And Enigmail is not compatible anymore. I try to do exactly was was explained in the thread https://portableapps.com/support/thunderbird_portable#encryption and when I load the add-on in TB, I have a message to tell me that enigmail v1.1 is not compatible with my version of TB.
So I would like to know if Enigmail is compatible with TB 3.0.4 portableapps?
For information. I directly updated TB and GPG portable ON the EXISTING TB and GPG portable.

On TB304, enigmail 1.0.1 works perfect.
You can backup your keys and reinstall the current version enigmail from the addons repository.
If some extension is not compatible, update the extension, restart TB this should fix all.
