It's always automatic for every new TB version. At any given time, the version if Lightning will work with TB X but not TB X+1. So as you upgrade, Lightning is disabled and then updated right after TB is. It's all automated.
Sometimes, the impossible can become possible, if you're awesome!
Sometimes the automatic update fails and you need to do a 'manual' update via add-ons > check for updates .
99% of the time if the automatic update during upgrade fails.
Then add-ons > check for updates will catch it and install the update.
Mozilla won't release a new Thunderbird without also updating Lightning.

I've always had a hard time finding updated versions of lightning.
The version here has not changed for some time:

and the one on this site hasn't changed for awhile either.

Was able to update the plugin via thunderbird during the upgrade process.
