I have just launched Thunderbird and can see this: https://i.imgur.com/2lqeMQl.png.
In settings I have it switched to: "Check for updates, but let me choose whether to install them".
Shouldn't update feature be totally disabled for portable application? I suspect an inexperienced user may launch auto-updates which in result will ruin portability of app.

The setting to disable updates was removed from Thunderbird itself. You can change it with a policy file now if you want. We're required by our licensing agreement with Mozilla to have updates on by default. Not automatic, but on in the fashion you see them. You can run Thunderbird's updater and it will update the currently running copy (32-bit or 64-bit) within Thunderbird Portable correctly and remain portable. Same with Firefox. Many people mainly use it this way. It won't update the other copy or portable launcher, though, so it's recommended to use our update process at least regularly.

There must be a way to prevent this?

The app has been updated so you can update manually or via the PA.c Platform. The delay for this release was due to handling the changed updater.
As stated above, you can manually fully disable the updater using a policies.json distribution file within the App\Thunderbird\distribution and/or App\Thunderbird64\distribution folder. We can not distribute Thunderbird or Firefox with this set due to licensing.
