This is incorrect. All my Papps are loaded on a USB microSD card which is working fine. I checked I can create a text file with Explorer in Firefox Portable's folder.

The launcher attempts to create a new file in your Profile directory called writetest.temp. If it's unable to open the file for writing, it shows the error you saw. It's likely there's some corruption, bad permissions on that particular directory, it's set to read only, etc.
Sometimes, the impossible can become possible, if you're awesome!

Sorry to bump up this topic, did you find a solution to the issue?
I experienced a similar issue due to ESET Browser Protection being enabled for all browsers.
After the issue with the weird prompts for permission from an administrator and subsequently from myself, Firefox Portable eventually ended up complaining about running from a read-only location.
If you are using ESET antivirus, you may find the solution under the topic named "Read-only location - Profile folder locked - ESET Browser Protection".
As a last resource, you may try to restart Windows under Safe Mode, and check if the issue persists, because there may be some other software interfering with Firefox functionality under Normal Mode.
So I am having same problem here with FirefoxPortable v143.0.1 on Windows 10 pro. After install I can open app only one time and then I get information that "firefox portable edition appears to be running from a location that is read-only"

What I have done untill now:
- unmarked read-only attribute for FirefoxPortable folder
- unmarked read-only attribute for folder %APPDATA%\Mozilla
- downloaded other language variants of FirefoxPortable
- tried to "install" FF Portable in different locations
I also tried to uninstall KB5065429 which I installed on my Win10 wright before FF stopped working, but it is impossible without a full system reinstall (no uninstall button or impossible with "wusa" Powershell command).
Any help?

Stopping ESET AV protection and firewall will not work. Only UNINSTALLING ESET "solved" this problem.

You do not have to install or uninstall anything.
The problem lies in the interaction of ESET and Firefox (and probably other browsers), thus there is absolutely no reason to make any adjustments to Windows, trying to install or uninstall specific updates.
Also, there is no point to try to change folder permissions, since the problem is not that you do not have the necessary permissions, but that the folders are being actively locked by the ESET services, which are still running even if you temporarily pause ESET protection or ESET firewall.
Last, but not least, you do not have to uninstall ESET, which in my opinion is an excellent antivirus program, you can leave it running as usual, and also leave enabled the generic ESET Browser Protection, which you can use for secure browsing.
To solve all these issues, you have to disable ESET Browser Protection for all browsers, which is the second option underneath the Enable Safe Banking & Browsing option in the ESET configurationsettings.
