You can not "assign" a drive letter to a usb drive and have it be the same on every pc, windows has no feature or function for that.
You can however run a script to substitute a drive letter when you plug the drive in.
Google subst.exe.
Or you can use my Menu or StartUp Util

What I did, was on all machines you have admin rights to, you go into Administrative Tools -> Computer Management -> Disk Management and changed the letter of the usb drive to always be J. Once its set the machine will always remember it. Since I primarily use my portable apps between three machines I own (deaktop, laptop, server) and I use CryptableApps and need admin rights anyway, its no big deal for me to have admin rights. I actually have a 2nd thumb drive I have a different setup of portable apps on, that I use at work, where I do not have admin rights.

As far as I've been able to determine, (and somebody correct me if I'm wrong) Windows by default automatically uses the next available drive letter.
